Smoky Moore rides into Pecos City to visit his adopted father, Sheriff Bill Hastings, for the first time in fifteen years. Before there can be a happy reunion, Smoky foils a midnight bank robbery. To his astonishment, the man behind the theft is the town's mayor. Jailed alongside Hastings, Smoky discovers that the city officials have been replaced by outlaws. With only hours before he will be hanged, Smoky calls on his old partners, The Lone Rider and Fuzzy Q. Jones, to come to his rescue. Smashing the crooked government of Pecos City will be the greatest challenge the three friends have ever faced when they find themselves the only good guys in a town of deranged killers.

The Lone Rider in Cheyenne is the eighth film in PRC's Lone Rider series. Beginning with The Lone Rider Rides Again in 1941, PRC produced 11 Lone Rider features starring baritone-voiced George Houston as the titular hero and Al St. John as his sidekick Fuzzy. Starting with the sixth entry, the studio brought in handsome Dennis Moore as the Rider's partner, rounding out the trio. Houston was replaced for the final six films by Bob Livingston. The Lone Rider in Cheyenne also features a memorable bad guy role for Roy Barcroft. The following year, Barcroft would sign an exclusive contract with Republic, where he would achieve immortal status as the studio's chief villain.
